



Coomassie® Brilliant Blue G 250 is a triphenylmethane dye commonly used in protein gel electrophoresis for the detection of proteins and in the Bradford method for determining protein concentration. It is widely utilized in laboratory applications requiring sensitive protein analysis and quantification.
Coomassie Brilliant Blue G 250 differs from the R 250 variant by the addition of two methyl groups and a slightly greenish tint to its blue coloration, providing distinct staining characteristics in protein assays.
The dye exists in multiple forms depending on pH conditions: a cationic (red) form below pH 0 with an absorbance peak at 470 nm, a neutral (green) form around pH 1 with an absorbance peak near 650 nm, and an anionic (blue) form above pH 2 with an absorbance peak at 595 nm. The blue anionic form binds to amino acid residues, including arginine and aromatic residues, forming a stable protein-dye complex.
