SERVA HPE™ Lightning Red Kit is a highly sensitive fluorescent labelling kit for rapid pre-labelling of proteins prior to 2D PAGE. It enables direct detection of proteins, eliminating the need for staining and washing steps after electrophoresis.
The dye is fully compatible with mass spectrometry and downstream applications such as Western blotting.
The labelling procedure is simple and fast: typically 80 pmol SERVA HPE™™ Lightning Red is used for labelling 1 µg of protein, followed by incubation at 0 °C for 15 minutes. The labelled protein solution can then be directly applied to an IPG strip via cup-loading or rehydration loading.
After the 2D run, detection is performed using a fluorescent imager (camera or scanner) with an excitation wavelength of approximately 530 nm and an emission filter of 610 nm (narrow band width of 30 nm).
The dye is compatible with common sample solubilization and extraction additives, including carrier ampholytes and reductants. Alkalescent conditions are sufficient, and pH adjustment to a defined value is not required.
The kit contains 250 µg of SERVA HPE™™ Lightning Red dye and DMSO.
